Description

TACI (transmembrane activator and calcium modulator and cyclophilin ligand interactor), a type I membrane protein, is a member of the TNF receptor superfamily. TACI interacts with two ligands, the BAFF (B cell activation factor) and APRIL (proliferation-inducing ligand). Engagement of TACI with its ligand(s) is thought to trigger inhibitory signal(s) as TACI-deficient mice develop fatal autoimmune glomerulonephritis, proteinurea, and elevated levels of circulating autoantibodies.

Treatment of B cells with TACI agonistic antibodies inhibits proliferation in vitro and activation of a chimeric receptor containing the TACI intracellular domain induces apoptosis. Interestingly, TACI mutations are associated with immunodeficiency in humans, as a significant proportion of CVID patients have TACI mutations. People with this condition produce abnormally low amounts of antibodies, which are needed for protection against infections.

In humans, the gene encoding this protein is located within the Smith-Magenis syndrome region on chromosome 17. Soluble TACI protein, particularly the dimeric protein fused with IgG Fc domain is a robust decoy for both BAFF and APRIL.
