CD27 is found on the surface of majority of blood T cells, both on the CD4 and CD8 subsets and is defined by a set of monoclonal antibodies. Antigen-activated T cells up-regulate the CD27 expression and CD27 interaction with its receptor augments Ag-receptor mediated T cell proliferation. Cloning of CD27 was accomplished by expression cloning using anti-CD27 mAbs from a human PBL cDNA library.
The CD27 protein extracellular domain contains 2 CRDs that exhibit similar sub-domain motifs to those of the CD40 and TNFR2. Similar to human NFGR, CD27’s membrane proximal region is over-represented by Pro, Ser and Thr amino acid residues which are often the O-linked glycosylation sites. This is confirmed by its slower mobility on SDS-PAGE and a shift of isoelectric point (pI) that would be otherwise predicted. CD27’s intracellular domain is relative short and does not have a death domain (DD). However, it interact with SIVA which does contains a DD. Overexpression of SIV triggers cell death suggesting potential role of SIVA in mediating CD27 transduced apoptotic signal.
