Description
The TRAIL-R4, also known as DcR2, was identified by hybridization to a probe based on the extracellular domain of TRAIL-R3. Together with DR4, DR5 and TRAIL-R3, the gene coding for TRAIL-R4 is mapped to human chromosome 8p21.
Similar to DR5, and possibly DR4, TRAIL-R4 has an unusually long signal peptide of 55 amino acids. TRAIL-R4 contains the canonical cysteine-rich domains and a truncated death domain, each is highly homologous to the respective domain of the DR4 and DR5, much less so to TNFR1, Fas and DR3. In contrast to TRAIL-R3, another decoy receptor for TRAIL, TRAIL-R4 does not contain the TAPE repeats and therefore its ligand binding site is on the same plane as that of the DR4 and DR5. While its partial death domain is clearly functionally compromised, TRAIL-R4 was shown to interact with DR4 and led to inhibition of TRAIL-induced apoptosis.
It is not completely clear that this inhibitory effect is resulting from the hetero-dimerization of TRAIL-R4 with DR4 via the pre-ligand assembly domain (PLAD) or from the simultaneous bindings of both receptors to a single TRAIL molecule that led to formation of non-functional DISC (death inducing signalling complex.
